Background of CCR5
CD195 / CCR5 (also known as CKR-5) is a receptor for inflammatory CC-chemokines (characterized by a pair of adjacent cysteine residues), such as MIP-1 alpha, MIP-1 beta, or RANTES. It is a G protein-associated seven-pass transmembrane protein expressed on resting T cells with memory/effector phenotype, monocytes, macrophages and immature dendritic cells. This chemokine receptor regulates the activation and directed migration of leukocytes. Importantly, along with CD4, CD195 / CCR5 functions as a major receptor for HIV. Their ligand is the viral glycoprotein gp120.

Anti-Hu CD195 purified antibody (clone T21/8) specificity verification by flow cytometry
Anti-Hu CD195 purified (concentration in sample 5 µg/ml, GAM PE, red-filled histogram) binds specifically to surface CD195 in transfected cells (upper panel), but not to the parent HEK-293 WT cells (lower panel).
Level of non-specific binding was assessed on cells unstained by primary antibody (GAM PE, black-dashed histogram).

Anti-Hu CD195 Purified (clone T21/8) works in WB application in non-reduced and non-boiled LM lysates.
Western blotting analysis was performed on 1% lauryl maltoside (LM) whole cell extracts of HEK293T/CD195 transfectants and HEK293T cells, mixed with non-reducing SDS-loading buffer without heating. Samples were resolved using 10% SDS-PAGE gel.
Nitrocellulose membrane blot was probed with mouse IgG1 monoclonal antibody T21/8 (1 µg/ml), followed by IRDye 800CW Goat-anti-Mouse IgG (green). Mouse anti-GAPDH monoclonal antibody FF26A conjugated with DyLight 680 (0.1 µg/ml) was used as the loading control (red). Multiplex fluorescent Western blot detection was performed.
CD195 monomers were detected at ~48 kDa in HEK293T/CD195 transfectants.
