Anti-JAB1 Monoclonal Antibody (Boster Biological Technology, Pleasanton CA, USA, Catalog # M01849)
Host
Rabbit
Contents
Rabbit IgG in stabilizing components, phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.
*This antibody is supplied in a stabilized formulation.
Compatibility with conjugation reactions depends on the chemistry of the conjugation method used.
For conjugation methods that are not compatible with the stabilizing components present in this formulation, a carrier-free antibody format is required.
Clonality
Monoclonal
Clone Number
AEEG-3
Isotype
Rabbit IgG
Immunogen
A synthesized peptide derived from human JAB1 Probable protease subunit of the COP9 signalosome complex (CSN), a complex involved in various cellular and developmental processes. The CSN complex is an essential regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of the cullin subunits of the SCF-type E3 ligase complexes, leading to decrease the Ubl ligase activity of SCF-type complexes such as SCF, CSA or DDB2.
Reactive Species
M01849 is reactive to COPS5 in Human, Mouse, Rat
Observed Molecular Weight
35 kDa
Calculated molecular weight
37.6 kDa
Antibody Validation
Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Western blot analysis of JAB1 using anti-JAB1 antibody (M01849).
Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human MCF-7 whole cell lysates,
Lane 2: human Jurkat whole cell lysates,
Lane 3: human U-87MG whole cell lysates,
Lane 4: human HepG2 whole cell lysates,
Lane 5: rat spleen tissue lysates,
Lane 6: rat lung tissue lysates,
Lane 7: mouse spleen tissue lysates,
Lane 8: mouse lung tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-JAB1 antigen affinity purified monoclonal antibody (M01849) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for JAB1 at approximately 35 kDa. The expected band size for JAB1 is at 38 kDa

Figure 5. IHC analysis of JAB1/COPS5 using anti-JAB1/COPS5 antibody (M01849).
JAB1/COPS5 was detected in a paraffin-embedded section of human breast cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with a dilution of 1:50 rabbit anti-JAB1/COPS5 Antibody (M01849) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.
