Background of KCNK12
KCNK13 and KCNK12 (also known as THIK1 and 2) are the first two members of a novel two pore-forming P domains K+ channels subfamily. The pore loop domain, a highly conserved region common to all potassium channels, is involved in determining potassium ion selectivity. Members of this family are all characterized by four transmembrane domains and may function to help influence the resting membrane potential of cells. KCNK12 is expressed mainly in the brain and lung, but also observed in the kidneys, heart and skeletal muscle. KCNK12 is closely related to KCNK13 (58% identity at the amino acid level), but could not yet been functionally expressed in vitro and may require other proteins to become active.

Assay Dilutions Recommendation
The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.
KCNK12 antibody can be used for detection of KCNK12 by Western blot at 0.5 μg/mL. Antibody can also be used for immunohistochemistry starting at 5 μg/mL. For immunofluorescence start at 20 μg/mL.
Antibody validated: Western Blot in rat samples; Immunohistochemistry in mouse samples and Immunofluorescence in mouse samples. All other applications and species not yet tested. Optimal dilutions for each application should be determined by the researcher.
