Background of GRM5
Metabotropic glutamate receptor 5?is a?G protein-coupled receptor?that in humans is encoded by the?GRM5 gene. It is mapped to 11q14.2-q14.3. This gene encodes a member of the G-protein coupled receptor 3 protein family. The encoded protein is a metabatropic glutamate receptor, whose signaling activates a phosphatidylinositol-calcium second messenger system. This protein may be involved in the regulation of neural network activity and synaptic plasticity. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. A pseudogene of this gene has been defined on chromosome 11. Alternative splicing results in multiple transcript variants.

Western blot analysis of Metabotropic Glutamate Receptor 5/GRM5 using anti-Metabotropic Glutamate Receptor 5/GRM5 antibody (A01338-3).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human U-87MG whole cell lysates,
Lane 2: human 293T whole cell lysates,
Lane 3: rat C6 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Metabotropic Glutamate Receptor 5/GRM5 antigen affinity purified polyclonal antibody (Catalog # A01338-3) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Metabotropic Glutamate Receptor 5/GRM5 at approximately 178 kDa. The expected band size for Metabotropic Glutamate Receptor 5/GRM5 is at 132 kDa.
