Background of Neuromedin-U
Neuromedin U (NMU) is a neuropeptide with potent activity on smooth muscle, which was isolated first from porcine spinal cord and later from other species. It is a member of the neuromedin family of neuropeptides. The encoded protein is a precursor that is proteolytically processed to generate a biologically active neuropeptide that plays a role in pain, stress, immune-mediated inflammatory diseases and feeding regulation. Increased expression of this gene was observed in renal, pancreatic and lung cancers. Alternative splicing results in multiple transcript variants encoding different isoforms. Some of these isoforms may undergo similar processing to generate the mature peptide.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat kidney tissue lysates,
Lane 2: mouse kidney tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NMU antigen affinity purified polyclonal antibody (Catalog # A01919-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for NMU at approximately 26KD. The expected band size for NMU is at 20KD.

We are currently using anti-NMU antibody A01919-1 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it likely that the antibody can work on bovine tissues as well?

The anti-NMU antibody (A01919-1) has not been tested for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity.
