Background of Ornithine Decarboxylase

Recognizes a 53kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Assay Dilutions Recommendation

The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.

Flow Cytometry (0.5-1ug/million cells in 0.1ml)
Immunofluorescence (1-2ug/ml)
Western Blot (1-2ug/ml)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT)
(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)
Optimal dilution for a specific application should be determined.

Asked: 2019-08-09

Answer

You can see on the product datasheet, M03138 anti-Ornithine Decarboxylase-1 (ODC-1) Monoclonal antibody as been validated on IF. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Would anti-Ornithine Decarboxylase-1 (ODC-1) Monoclonal antibody M03138 work on feline IF with esophagus testis?

B. Miller

Verified customer

Asked: 2016-12-14

Answer

Our lab technicians have not tested anti-Ornithine Decarboxylase-1 (ODC-1) Monoclonal antibody M03138 on feline. You can run a BLAST between feline and the immunogen sequence of anti-Ornithine Decarboxylase-1 (ODC-1) Monoclonal antibody M03138 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline esophagus testis in IF, you can get your next antibody for free.
