Background of PER2

The PER2 gene encodes the period circadian protein homolog 2 protein in humans. This gene is a member of the Period family of genes and is expressed in a circadian pattern in the suprachiasmatic nucleus, the primary circadian pacemaker in the mammalian brain. PER2 is mapped to 2q37.3. It has been showed that PER2 can positively stimulate activity of the Bmal1-Npas2 transcription complex and, in turn, Npas2 transcriptionally regulates delta-aminolevulinate synthase. PER2 is involved in a negative-feedback loop that attenuates estrogen stimulation and links the circadian cycle to estrogen receptor signaling. The rhythmic levels of PER2 are critical for circadian oscillations in cells and intact organisms. What’s more, the PER2 gene also functions in tumor suppression by regulating DNA damage-responsive pathways.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Question

We are currently using anti-PER2 antibody PB9312 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on zebrafish tissues as well?

Verified Customer

Verified customer

Asked: 2019-10-02

Answer

The anti-PER2 antibody (PB9312) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in zebrafish you can get your next antibody for free. Please contact me if I can help you with anything.

Question

Does PB9312 anti-PER2 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2017-06-15

Answer

It shows on the product datasheet, PB9312 anti-PER2 antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
