Background of PIAS4
E3 SUMO-protein ligase PIAS4, also known as protein inhibitor of activated STAT protein 4 (PIAS4) or protein inhibitor of activated STAT protein gamma (PIASg or PIASy), is an enzyme that in humans is encoded by the PIAS4 gene. This gene is mapped to 19p13.3. This gene plays a crucial role as a transcriptional coregulation in various cellular pathways, including the STAT pathway, the p53/TP53 pathway, the Wnt pathway and the steroid hormone signaling pathway. It also functions as an E3-type small ubiquitin-like modifier (SUMO) ligase, stabilizing the interaction between UBE2I and the substrate, and as a SUMO-tethering factor. This gene involved in gene silencing.

Western blot analysis of PIAS4 using anti-PIAS4 antibody (PB10082).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat testis tissue lysates,
Lane 2: mouse testis tissue lysates,
Lane 3: HELA whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PIAS4 antigen affinity purified polyclonal antibody (Catalog # PB10082) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PIAS4 at approximately 57 kDa. The expected band size for PIAS4 is at 57 kDa.

The anti-E3 SUMO-protein ligase PIAS4 antibody (PB10082) has not been validated for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. Answer
You can see on the product datasheet, PB10082 anti-E3 SUMO-protein ligase PIAS4 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
