Background of PLA2G4A
PLA2G4A (Phospholipase A2, Group IVA), is an enzyme that in humans is encoded by the PLA2G4A gene. Tay et al. (1995) mapped the PLA2G4A gene to rat chromosome 13 by PCR-based intercross genotyping and to human 1q25 by fluorescence in situ hybridization. By site-directed mutagenesis and biochemical analysis of the recombinant protein, Sharp et al. (1994) determined that ser228 participates in the catalytic mechanism of cPLA2 and that both the phospholipase A2 and the lysophospholipase activities are catalyzed by the same active site residue (s). PLA2G4A, the cytosolic phospholipase A2, appears to subserve transmembrane signaling responses to extracellular ligands (Skorecki, 1995).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 40 ug of sample under reducing conditions.
Lane 1: HELA Whole Cell Lysate,
Lane 2: NIH/3T3 Whole Cell Lysate.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PLA2G4A antigen affinity purified polyclonal antibody (Catalog # PB9778) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PLA2G4A at approximately 85 kDa. The expected band size for PLA2G4A is at 85 kDa.

We are currently using anti-Phospholipase A2/PLA2G4A antibody PB9778 for human tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on pig tissues as well?

The anti-Phospholipase A2/PLA2G4A antibody (PB9778) has not been validated for cross reactivity specifically with pig tissues, but there is a good chance of cross reactivity.
