Description
PROK1 (prokineticin-1; EG-VEGF) is a secreted factor reported to induce proliferation, migration, and fenestration in endothelial cells and to potently contract GI smooth muscle, supporting endocrine-tissue microvascular and smooth-muscle signaling contexts. Assay context: human-reactive antibody tested for capture ELISA and for protein confirmation by Western blot (per provided product description); pairs naturally with the corresponding quantitative kit PROK1 (EG-VEGF) ELISA when aligning immunoreactivity with soluble analyte levels (putative workflow linkage).
Storage & Handling
Store at -20˚C for one year from date of receipt. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for six months. Avoid repeated freeze-thaw cycles.

Background of PROK1
Prokineticin-1 is also known as EG-VEGF. The International Radiation Hybrid Mapping Consortium mapped the PROK1 gene to chromosome 1. The protein encoded by this gene induces proliferation, migration, and fenestration (the formation of membrane discontinuities) in capillary endothelial cells derived from endocrine glands. It has little or no effect on a variety of other endothelial and non-endothelial cell types. Its expression is restricted to the steroidogenic glands (ovary, testis, adrenal, and placenta), is induced by hypoxia, and often complementary to the expression of vascular endothelial growth factor (VEGF), suggesting that these molecules function in a coordinated manner.

Western blot analysis of Prokineticin 1 using anti-Prokineticin 1 antibody (PB9881).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: MCF-7 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Prokineticin 1 antigen affinity purified polyclonal antibody (Catalog # PB9881) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Prokineticin 1 at approximately 20 kDa. The expected band size for Prokineticin 1 is at 12 kDa.
