Western blot analysis of Pygopus 2 using anti-Pygopus 2 antibody (M00283).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human MCF-7 whole cell lysates,
Lane 3: human SH-SY5Y whole cell lysates,
Lane 4: human PC-3 whole cell lysates,
Lane 5: huamn U2OS whole cell lysates,
Lane 6: mouse NIH/3T3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Pygopus 2 antigen affinity purified monoclonal antibody (M00283) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:500 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Pygopus 2 at approximately 50 kDa. The expected band size for Pygopus 2 is at 41 kDa.

Question
Would anti-Pygopus 2 Rabbit Monoclonal antibody M05260 work on dog WB with nasal cavity epithelium?
Verified Customer
Verified customer
Asked: 2018-07-16
Answer
Our lab technicians have not tested anti-Pygopus 2 Rabbit Monoclonal antibody M05260 on dog. You can run a BLAST between dog and the immunogen sequence of anti-Pygopus 2 Rabbit Monoclonal antibody M05260 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated dog samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in dog nasal cavity epithelium in WB, you can get your next antibody for free.
