Description
SGCA (α-sarcoglycan) is a core component of the sarcoglycan complex within the dystrophin-associated glycoprotein complex (DGC), supporting skeletal/cardiac muscle membrane stability; loss-of-function is linked to sarcoglycanopathies and membrane fragility–associated injury (canonical). Assay context: rabbit anti-SGCA primary validated for Western blot with Human/Mouse/Rat reactivity; can be used to contextualize muscle injury/remodeling readouts such as extracellular matrix turnover (putative), including collagen I production measured by COL1A1 ELISA."
Storage & Handling
Store at -20˚C for one year from date of receipt. After reconstitution, at 4˚C for one month. It can also be aliquotted and stored frozen at -20˚C for six months. Avoid repeated freeze-thaw cycles.

Background of SGCA
Alpha-sarcoglycan is a protein that in humans is encoded by the SGCA gene. This gene encodes a component of the dystrophin-glycoprotein complex (DGC), which is critical to the stability of muscle fiber membranes and to the linking of the actin cytoskeleton to the extracellular matrix. Its expression is thought to be restricted to striated muscle. Mutations in this gene result in type 2D autosomal recessive limb-girdle muscular dystrophy. Multiple transcript variants encoding different isoforms have been found for this gene.

Western blot analysis of SGCA using anti-SGCA antibody (PB9503).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Skeletal Muscle Tissue Lysate at 50ug,
Lane 2: Mouse Skeletal Muscle Tissue Lysate at 50ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SGCA antigen affinity purified polyclonal antibody (Catalog # PB9503) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SGCA at approximately 43 kDa. The expected band size for SGCA is at 43 kDa.

We are currently using anti-alpha Sarcoglycan/SGCA antibody PB9503 for mouse tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on horse tissues as well?

The anti-alpha Sarcoglycan/SGCA antibody (PB9503) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
