What Is the Expected LPCAT1 Western Blot Band Size?

LPCAT1 is predicted at 59.2 kDa and observed at approximately 59 kDa; the small difference has no established cause in the supplied evidence.

What am I looking at on my blot?
Band at approximately 59 kDaMatches the empirical LPCAT1 band and its 59.2 kDa predicted mass
Single sharp band near 59 kDaConsistent with the predicted size and absence of listed size-modifying features
Weak band near 59 kDa in whole-cell lysateMembrane-associated LPCAT1 may be incompletely recovered
Band near 59 kDa in a membrane fractionConsistent with LPCAT1 membrane localization
💡Expected LPCAT1 appearanceLPCAT1 has a predicted mass of 59.2 kDa and an empirical band at approximately 59 kDa; confirm identity with appropriate antibody and sample controls.
How each factor affects band size
UniProt predicted molecular weightPlaces full-length LPCAT1 near 59.2 kDa
UniProt mass of 59,151 DaCorresponds to approximately 59.2 kDa
Predicted mass versus empirical bandThe 59.2 kDa prediction closely matches the approximately 59 kDa observed band
Predicted mass as band referenceBands far from 59.2 kDa require independent identity checks
Why is my band missing or off?

Sample controls for LPCAT1 Western blot

🧪HPA-IHC candidate guidance (verify in WB): For positive controls for LPCAT1 in Western blot, you can use bronchus tissue, which shows high HPA expression.
Positive control: Bronchus (IHC candidate; verify WB)
Negative control: Adipose tissue (IHC candidate; verify WB)
Loading controls: Run GAPDH, β-actin, and a total-protein stain alongside the samples.
⚠️Feasibility: LPCAT1 is membrane-associated, so use lysates that retain membrane proteins.

Advanced LPCAT1 Western Blot Tips

Deeper troubleshooting and optimisation questions for LPCAT1, answered from its protein features.

How should LPCAT1 band migration be interpreted?
Band shift · Use the separately labelled calculated mass and catalog-observed evidence above. A sequence annotation does not establish an observed migration shift. Verify target identity with orthogonal controls.
Could annotated isoforms explain multiple LPCAT1 bands?
Isoforms · Only one isoform is supplied, with no alternative sequence. The supplied features therefore do not identify an isoform that explains an additional band.
Do annotated modifications explain a shifted LPCAT1 band?
PTM · No modified residues or glycosylation sites are supplied. The supplied features do not support assigning a shifted band to a specific modification.
Does this guide establish induction of LPCAT1?
Induction · No general induction response is established by this guide. A pathway or gene-set association is not evidence of induction in a particular specimen. Verify the relevant treatment and control in a target-specific experiment.
What transfer method to use for LPCAT1 Western blot?
Transfer · LPCAT1 is annotated as a 59.2 kDa single-pass membrane protein. These features do not establish one best transfer method. Check transfer efficiency near 59 kDa when setting up the blot.
How should blocking be optimized?
Blocking · Standard workflow guidance: follow the A04863-1 datasheet where specified. Otherwise compare 5% milk or 5% BSA in TBST; for a phospho-specific assay start with BSA. Optimize background and specific signal with matched controls.
How should LPCAT1 bands be quantified?
Quantitation · Quantify the band near the supplied observed position of 59 kDa, using the same band definition across samples. The features alone do not establish that other bands represent LPCAT1.
Should LPCAT1 migrate above its predicted mass?
Interpretation · The predicted mass is 59.2 kDa, and the supplied observed band is about 59 kDa. The features do not establish a reason to expect a shift.

Compare it with the approximately 59 kDa observed band. One isoform is supplied, and no modified residues or glycosylation sites are annotated, so the supplied features do not identify the cause of an additional band.

LPCAT1 Western Blot Antibodies

Catalog antibodies with Western blot application and product-specific WB images. Evaluate suitability with the reported sample, controls and experimental conditions.

Real WB data Western blot analysis of LPCAT1 using anti-LPCAT1 antibody (A04863-1). Electrophoresis was performed on a 10% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human A431 whole cell lysates, Lane 2: human A549 whole cell lysates, Lane 3: human PC-3 whole cell lysates, Lane 4: human Hela whole cell lysates, Lane 5: rat lung tissue lysates, Lane 6: mouse lung tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-LPCAT1 antigen affinity purified polyclonal antibody (A04863-1) at 1: 1000 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an ECL Plus Western Blotting Substrate (Catalog # AR1196-200) with Tanon 5200 system. A specific band was detected for LPCAT1 at approximately 59 kDa. The expected band size for LPCAT1 is at 59 kDa.

Two the supplier anti-LPCAT1 antibodies, A04863-1 and A04863, list Human, Mouse, and Rat reactivity and include Western blot images. A04863-1 documents named human cell and rat and mouse lung lysates; A04863 describes only ‘various lysates,’ limiting sample-specific interpretation.

Which to pick: Both list the same species and have WB images. Choose A04863-1 when its documented A431, A549, PC-3, HeLa, rat lung, or mouse lung conditions match your experiment; its caption reports a band near 59 kDa. A04863 gives fewer sample details.
