What Is the Expected MDM4 Staining Pattern?

MDM4 is mainly nuclear across tissues in HPA IHC, with high staining reported in several epithelial, glandular, neuronal and stromal cell groups (HPA tissue IHC). UniProt also lists cytoplasmic localisation and no transmembrane segment (UniProt O15151). HPA rates the tissue IHC pattern Approved but reports low consistency with RNA expression and a splice or transcript discrepancy; interpret intensity with that caution (HPA tissue IHC).

What am I looking at on my slide?
Predominant nuclear chromogen in adrenal glandular cells or cortical neurons. This fits HPA's mainly nuclear profile and its High staining calls for those cells (HPA tissue IHC). Score the stained nuclei and their cell identity together; a positive field alone does not establish antibody specificity, given HPA's RNA and staining consistency warning (HPA tissue IHC).
Predominantly membranous, extracellular or uniformly cytoplasmic staining with little nuclear signal.Treat this as a discordant IHC pattern: HPA reports mainly nuclear tissue staining, while UniProt lists nucleus and cytoplasm and no transmembrane segment (HPA tissue IHC; UniProt O15151). Cytoplasmic signal is possible, but a dominant nonnuclear pattern needs scrutiny before scoring it as specific.
Strong staining concentrated in an unexpected cell population while expected nuclei remain weak.Compare cell identities within the same section. HPA reports Low staining in duodenal glandular, urothelial, smooth muscle and marrow hematopoietic cells, but does not call them negative (HPA tissue IHC). A strong reversal raises possible cross-reactivity or endogenous detection activity; morphology alone cannot distinguish those causes.
Diffuse chromogen over cells and extracellular spaces, with little nuclear contrast. This obscures the mainly nuclear pattern expected from HPA tissue IHC (HPA tissue IHC). In general IHC practice, broad background can arise from nonspecific antibody binding, incomplete blocking or detection chemistry. Do not convert diffuse colour into a positive MDM4 score without a cell-resolved nuclear pattern.
No nuclear signal in an adrenal gland or bronchus section chosen as a positive comparator.HPA reports High staining in adrenal glandular and bronchial respiratory epithelial cells (HPA tissue IHC). Check whether those cells are present and whether a parallel control stained. A blank result may reflect the assay or section; HPA's Approved rating and consistency warning do not guarantee every specimen will stain.
💡Expected MDM4 appearanceCall a convincing IHC positive when identifiable cells show predominantly nuclear chromogen, with potentially strong staining in an HPA High cell group; diffuse extracellular colour or a dominant membrane outline is discordant (HPA tissue IHC; UniProt O15151 topology).
How each factor affects the staining
Tissue and cell selectionHPA reports High staining in adrenal glandular, bronchial respiratory epithelial, cortical neuronal and endometrial stromal cells; several other listed cell groups are Low (HPA tissue IHC). Choose comparators by cell type and nuclear pattern, and avoid treating a Low call as an absent-protein control.
Evidence strengthThe tissue IHC profile is Approved, with low consistency between antibody staining and RNA expression and a splice or transcript discrepancy (HPA tissue IHC). The listed IHC antibody HPA018919 is Approved; HPA does not list it as Enhanced (HPA antibodies). Interpret unexpected intensity conservatively.
Localisation and topologyHPA describes mainly nuclear tissue staining; UniProt lists nuclear and cytoplasmic localisation and no transmembrane segment (HPA tissue IHC; UniProt O15151). A small cytoplasmic component need not invalidate nuclear staining, while a membrane-only pattern lacks support from these records.
Isoforms and processingUniProt lists five MDM4 isoforms, no signal peptide or propeptide, and a 1–490 protein chain (UniProt O15151). These facts support caution about transcript-dependent comparisons; the payload gives no antibody epitope map or evidence that a particular isoform changes IHC staining.
IF comparisonHPA ICC-IF supports nucleoplasmic localisation, while its tissue IHC profile says mainly nuclear (HPA subcellular; HPA tissue IHC). Use the IF finding as a compartment check only; the supplied records do not establish that IF intensity predicts paraffin-section chromogenic intensity.
Why is my staining missing, weak or wrong?
SituationLikely causeNext action
Expected High cell group is blank. The expected cells may be absent, or the staining run may have failed; HPA reports High staining for specific cells within listed tissues, not every cell in each section (HPA tissue IHC).Verify cell identity on the counterstain and inspect a concurrently stained control. Then review the assay's antigen retrieval, primary antibody and detection steps as general IHC checks; no MDM4-specific retrieval or fixation sensitivity is supplied.
Only a membrane outline is strong. That pattern conflicts with mainly nuclear HPA tissue staining and UniProt's lack of a transmembrane segment (HPA tissue IHC; UniProt O15151).Check localisation at higher magnification against the counterstain, then compare a control section. Treat membrane-only colour as unresolved staining rather than assigning it to MDM4.
Low-call cells stain more strongly than the chosen High-call cells.HPA's Low and High calls are observations with a stated RNA and staining consistency caveat (HPA tissue IHC). Cell misidentification, nonspecific binding or endogenous detection activity are possible general IHC explanations.Recheck both cell populations and the nuclear pattern; examine appropriate primary-omission and detection controls. Report the discrepancy instead of treating the HPA categories as absolute positive and negative thresholds.
Diffuse background hides nuclei.General IHC causes include nonspecific binding, inadequate blocking or excess detection signal; the supplied HPA and UniProt records do not identify an MDM4-specific background mechanism.Inspect primary-omission and detection controls, then review blocking, antibody concentration, washes and chromogen development under the assay's established conditions. Score MDM4 only where discrete nuclei remain interpretable.
Nuclear staining is present, but intensity differs across specimens.HPA reports broadly nuclear expression with cell-specific intensity calls and warns of low RNA and staining consistency plus a splice or transcript discrepancy (HPA tissue IHC).Compare the same cell type and compartment across sections and record intensity separately from localisation. Avoid inferring isoform identity or transcript abundance from chromogen intensity alone.
ICC-IF looks nucleoplasmic, but paraffin IHC is weak.HPA supports nucleoplasmic ICC-IF localisation and reports mainly nuclear tissue IHC, but these records do not establish equivalent signal strength across assays (HPA subcellular; HPA tissue IHC).Assess each assay with its own controls. For paraffin IHC, review the established retrieval and detection workflow as general practice; do not infer an MDM4-specific fixation effect from the ICC-IF result.

Sample controls for MDM4 IHC & IF

🧪Run bronchus first and score respiratory epithelial cells, which HPA rates High for MDM4 (HPA: bronchus, respiratory epithelial cells, High). HPA detects MDM4 in all 45 scored tissues, so there is no supported negative tissue; use no-primary and isotype controls, and expect cells without specific staining on the positive slide to show only background rather than treating any cell type as a validated internal negative (HPA: no negative rows; detected in all 45 scored tissues).
Positive control tissue: Adrenal gland (Glandular cells, HPA High)
Negative control tissue: None in HPA: MDM4 is detected in all 45 scored tissues. Use a no-primary (secondary-only) and an isotype control instead.
ICC-IF cell lines (HPA subcellular resource): HPA ICC-IF images show MDM4 in A-431, U-251MG, U2OS, NIH 3T3, with annotated localisation: Nucleoplasm (supported) (HPA subcellular).
Technical controls: Include a no-primary, secondary-only slide, a concentration-matched rabbit IgG isotype control matched to the primary antibody’s clonality, and a biological negative made with MDM4 knockout material or a validated peptide block (selected PB9872 caption: rabbit primary). Quench endogenous peroxidase and assess endogenous biotin background because the selected paraffin-section method uses biotinylated secondary antibody, streptavidin–biotin complex and DAB (selected PB9872 caption: SABC/DAB detection).
⚠️Feasibility: No target-specific fixation window or fixation effect is reported, and the selected PB9872 tissue-IHC caption does not state its fixative (selected PB9872 caption: fixative unreported). The caption uses heat retrieval in EDTA at pH 8.0, but does not establish that retrieval is required for MDM4 (selected PB9872 caption: EDTA heat retrieval); the supplied evidence does not establish whether frozen sections or IF are easier, although HPA reports supported nucleoplasmic localization in ICC-IF (HPA: nucleoplasm, supported). In bronchus, assess airway mucus and intrinsic tissue background when scoring epithelial staining (standard IHC practice).

Advanced MDM4 IHC Tips

Troubleshoot MDM4 staining in paraffin sections by checking retrieval, nuclear localisation, antibody epitope coverage and controls before comparing signal across samples.

What retrieval should I try first when MDM4 staining is weak?
Start with heat mediated retrieval in EDTA at pH 8.0 (datasheet PB9872). The catalog antibody produced chromogenic staining in a paraffin section after this retrieval, followed by 1 μg/ml primary antibody overnight at 4°C (caption PB9872). If staining remains weak, compare retrieval duration on adjacent sections while holding antibody concentration and detection conditions constant (standard IHC practice). Check that stronger staining appears predominantly in nuclei, since tissue staining is mainly nuclear and nucleoplasmic localisation is supported (HPA tissue IHC; HPA subcellular). Excess retrieval can damage morphology or raise background, so assess both signal and tissue preservation (standard IHC practice).
Could fixation explain weak or uneven MDM4 staining?
MDM4 specific sensitivity to fixation is unknown because the selected tissue caption does not state a fixative (caption PB9872). Record the fixative, fixation duration and section handling for each specimen, then compare matched sections processed together (standard IHC practice). Keep the documented EDTA retrieval at pH 8.0 and 1 μg/ml primary incubation constant while assessing whether fixation differences track with signal (datasheet PB9872; caption PB9872). Inspect nuclear detail and tissue integrity before increasing detection strength, since processing damage can complicate chromogenic scoring (standard IHC practice). Do not use the observed tissue staining pattern to infer a preferred fixative (HPA tissue IHC).
How should nuclear and cytoplasmic MDM4 staining be judged?
Expect predominantly nuclear tissue staining, with supported localisation to the nucleoplasm (HPA tissue IHC; HPA subcellular). Cytoplasmic localisation is also recorded, so a cytoplasmic signal requires assessment rather than automatic rejection (UniProt O15151 subcellular). Compare cytoplasmic intensity with nuclear staining in the same cells and with a no primary control under identical detection conditions (standard IHC practice). MDM4 has no transmembrane segment, so a crisp cell surface rim is inconsistent with its recorded topology and merits a background check (UniProt O15151 topology). Score nuclear and cytoplasmic compartments separately to preserve that distinction across specimens (standard IHC practice).
Could isoforms or phosphorylation change what my antibody detects?
MDM4 has 5 listed isoforms, so antibody staining depends on whether its epitope is present in each expressed form (UniProt O15151 isoforms; standard IHC practice). Check the catalog antibody's stated immunogen or epitope against the isoforms before treating variable staining as a change in total MDM4 (standard IHC practice). The protein includes a SWIB/MDM2 domain at residues 25–108 and reported phosphorylation at residues 342 and 367 (UniProt O15151 domains; UniProt O15151 modified residues). Without an epitope map or direct testing, neither isoform coverage nor phosphorylation sensitivity can be assigned to this antibody (standard IHC practice). Interpret discordant staining alongside the reported transcript discrepancy caution (HPA tissue IHC).
How can I check an MDM4 result with multiplex immunofluorescence?
For a separate IF assay, pair MDM4 with a marker identifying the cell population being assessed and use a nuclear counterstain to locate signal within cells (standard IF practice). Choose fluorophores whose emission is distinguishable from the specimen's autofluorescence, and include single stain and no primary controls when setting channels (standard IF practice). MDM4 is recorded in the nucleus and cytoplasm and has no transmembrane segment, so an intracellular epitope generally requires permeabilisation after fixation (UniProt O15151 subcellular; UniProt O15151 topology; standard IF practice). Optimise that step for the IF antibody and specimen, because the paraffin section caption supplies no IF fixation or permeabilisation conditions (caption PB9872).
What should I check when DAB appears outside expected MDM4 positive cells?
First compare a no primary section with the stained section to identify detection reagent or tissue derived background (standard IHC practice). The documented workflow used 10% goat serum, a biotinylated secondary for 30 minutes at 37°C, a streptavidin biotin complex and DAB (caption PB9872). Apply a peroxidase block as a general chromogenic IHC step and evaluate whether endogenous biotin contributes to staining in this detection system (standard IHC practice; caption PB9872). If background persists, compare blocking and washing conditions on adjacent sections while retaining the documented 1 μg/ml primary condition (standard IHC practice; caption PB9872). Treat diffuse staining without a convincing nuclear component cautiously (HPA tissue IHC).
How should I quantify MDM4 across differently stained sections? ⚠ ANSWER MARKED FOR VERIFICATION
Define the scored cell population and tissue area before measuring MDM4, then apply the same thresholds to every section (standard IHC practice). Record the percentage of positive nuclei and intensity, or calculate a nuclear H-score from intensity weighted percentages; report cytoplasmic staining separately (standard IHC practice; HPA tissue IHC). If counting positive cells, express density per mm² of evaluable tissue and exclude folds, necrosis and damaged edges consistently (standard IHC practice). Normalise comparisons to the number of eligible cells or evaluable area, and include a matched reference section across staining runs (standard IHC practice). Report the compartment because tissue staining is mainly nuclear (HPA tissue IHC).
When is an apparent MDM4 positive result likely to be artefact?
A credible result should show a cellular pattern consistent with mainly nuclear tissue staining and supported nucleoplasmic localisation (HPA tissue IHC; HPA subcellular). Compare the signal with no primary controls and examine whether it concentrates at section edges, folds or necrotic regions, where staining artefacts can occur (standard IHC practice). Investigate DAB signal in controls for endogenous peroxidase activity and consider endogenous biotin in the documented streptavidin biotin workflow (standard IHC practice; caption PB9872). A strong cell surface rim is difficult to reconcile with MDM4 lacking a transmembrane segment (UniProt O15151 topology). Interpret weak or unexpected cell patterns cautiously because antibody staining and RNA data have low consistency (HPA tissue IHC).
