Background of BMP2

Bone morphogenetic protein-2 (BMP-2, BMP 2A) belongs to the transforming growth factor beta (TGF-beta) superfamily. It is thought to be involved in cartilage and bone formation during embryogenesis, but may have additional functions in morphogenesis as implied by its expression in various organs and embryonic tissues of mice.1 BMP-2 has been identified as a candidate mediator of retinoid activity. BMP-2 protein induces medulloblastoma cell apoptosis, whereas the BMP-2 antagonist noggin blocks both retinoid and BMP-2-induced apoptosis. BMP-2 also induces p38 mitogen-activated protein kinase (MAPK), which is necessary for BMP-2 and retinoid-induced apoptosis.2 Bone morphogenic proteins (BMPs) are known to promote osteogenesis, and clinical trials are currently underway to evaluate the ability of certain BMPs to promote fracture-healing and spinal fusion.3 The standard product used in this kit is recombinant human BMP-2, constituting dimer by two chains of 114 amino acids with the molecular mass of 26KDa.

Our lab hasn't detected recombinant BMP-2 expressed in E.coli and we are uncertain if the kit is able to detect it. We suggest the customer run a pilot test to see if the kit will work.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test BMP2 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

Q: What is the optimal O.D. value for BMP2 ELISA kit? I used your BMP2 ELISA on serum samples. For my positive control, I received an O.D. value of 0.826, while my negative control received a value of 0.136. I obtained both of these controls from the ELISA kit, where your kit's typical data shows O.D. values much higher than my positive control and your background is lower. My samples O.D. values are around 0.225 and the highest is only 0.357. can I consider these samples contain BMP2 even though the O.D. values are not very high?

A: The absolute O.D. values may change according to incubation time. The more you incubate the higher the O.D. values are going to be. a point of focus should be is whether your sample O.D. values are statistically significantly higher than your blank values. regarding your assay, you could extend your development time in the substrate incubation step to obtain higher O.D. values, as long as your negative controls' O.D. values are not increasing faster in relation to your positive controls. normally, a sample with O.D. value 2 standard deviations higher than your negative controls can be considered positive. We calculate the sensitivity of this ELISA kit by changeing cutoff O.D. value, calculated as the average of 20 negative controls plus 2 standard deviations of the 20 negative controls, into a concentration. in other words, when we claim this BMP2 ELISA kit to have sensitivity of 2pg/ml, that means the minimum amount of BMP2 that can be declared/interpreted as positive by the above standard is 2pg/ml.

We do get a lot of feedback that BMP-2 has cross-reactivity with other cytokines. According to our test, the most cross-reactive target is BMP-4 (< 1%). However, there are some detectable cross-reactivities with the following: BMP-3, -5, -6 and -7 as well as other proteins in the TGF family, such as TGF Beta 1, 2 and 3. We have tested it against Activin and a few other families but no cross-reactivity was detected.

Answer

The standard in the kit is rhBMP2(CHO, Q283-R396). The kit can recognize properly folded active rhBMP2 homodimer and rhBMP2 monomer in theory, we are not clear about the other two. Our ELISA kit's coating antibody does recognize the specific sequence in the many forms of rhBMP2. However the affinity is dependent on the integrity of the tertiary structure as well as proper exposure of the sequence on the aggregate surface. If monomer and properly folded homodimer have the same local tertiary structure then our kit will be able to recognize both. However this can only be determined by a pilot experiment.
