Background of 4-1BB/TNFRSF9/CD137
CD137 is a member of the tumor necrosis factor (TNF) receptor family. Its alternative names are tumor necrosis factor receptor superfamily member 9 (TNFRSF9), 4-1BB and induced by lymphocyte activation (ILA). CD137 gene was localized to chromosome 1p36.Members of this receptor family and their structurally related ligands are important regulators of a wide variety of physiological processes and play an especially important role in the regulation of immune responses. CD137 can be expressed by activated T cells, but to a larger extent on CD8 than on CD4 T cells. The best characterized activity of CD137 is its costimulatory activity for activated T cells. Crosslinking of CD137 enhances T cell proliferation, IL-2 secretion survival and cytolytic activity. Further, it can enhance immune activity to eliminate tumors in mice.

A: for those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer may affect immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

I purchased Human TNFRSF9/4-1BB ELISA Kit and in my results showed unspecific binding band in my concentrated HEK293F cell supernatant, having a size of around 70kDa. What particular extracellular epitope should this antibody supposed to bind? Do this antibody binds to other TNFR superfamily proteins?

Answer
The Human TNFRSF9/4-1BB ELISA Kit immunogen is between aa to aa of human protein and this antibody should pick-up the target protein at 30-35kDa. Is the 70kDa band a dimer molecules? As the target protein usually form dimers. If not, we suggest to use beta-Mercaptoethanol or DTT with anionic denaturing detergent in your loading buffer. Also heat your sample at 95C in the reducing-denaturing buffer for 5mins up to 10mins if necessary.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test CD137 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

Q: we need your recommendation regarding the dilution ratio of serum samples for detection of CD137 in Human serum? I am trying to measure a few analytes and it requires 100ul of diluted samples for each well. We have limited sample quantitys so we like to dilute as much as possible.

A: unable to understand the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the CD137 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.
