Description
S100A9 is a calcium/zinc-binding S100 protein with a prominent role in inflammatory processes and immune responses; commonly studied in myeloid activation (often as part of S100A8/A9 “calprotectin” biology). Assay context: quantitative sandwich ELISA (96 tests) for mouse S100a9 in culture supernatants, cell lysates, serum, and plasma (heparin/EDTA), sensitivity 10 pg/mL; often interpreted alongside human innate-inflammatory counterparts such as S100A12 or chemokines like CCL4 in cross-species inflammation panels (putative).
Storage & Handling
Store at 4°C for 6 months, at -20°C for 12 months. Avoid multiple freeze-thaw cycles (Ships with gel ice, can store for up to 3 days in room temperature. Freeze upon receiving.)

Background of S100a9
Protein S100A9 also known as migration inhibitory factor-related protein 14 (MRP-14) or calgranulin-B is a protein that is encoded by the S100A9 gene. S100A9 is a member of the S100 family of proteins containing 2 EF hand calcium-binding motifs. It is mapped to Chromosome 3 in mouse. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. This protein may function in the inhibition of casein kinase. S100A9 complexes with S100A8, another member of the S100 family of calcium-modulated proteins regulates myeloid cell function by binding to Toll-like receptor-4 (TLR-4) and the receptor for advanced glycation end products.

A: Boster Bio does not evaluate sample stability. Variations in sample collection, processing, and storage may alter the stabilityof samples. It is recommend to assay sample right after collection when possible, or aliquot into single use volumes and store samples frozen. avoid repetitive freeze-thaw cycles with the stored samples to prevent protein degradation.

A: Chelating agents such as EDTA, Heparin and Citrate can bind metal ions from the functional domain of S100A9 causing disruption of its protein structure. S100A9 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes needed for detection and block the antigen antibody reaction. We have tested the S100A9 ELISA, treating samples with a number of anticoagulants and decided that heparin or EDTA can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test S100A9 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.
