Background of Perilipin-2/ADFP

Recognizes a protein of 48kDa, which is identified as Adipophilin. It belongs to the perilipin family, members of which coat intracellular lipid storage droplets. This protein is associated with the lipid globule surface membrane material, and maybe involved in development and maintenance of adipose tissue. However, it is not restricted to adipocytes as previously thought, but is found in a wide range of cultured cell lines, including fibroblasts, endothelial and epithelial cells, and tissues, such as lactating mammary gland, adrenal cortex, Sertoli and Leydig cells, and hepatocytes in alcoholic liver cirrhosis, suggesting that it may serve as a marker of lipid accumulation in diverse cell types and diseases.Â

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Assay Dilutions Recommendation

The recommendations below provide a starting point for assay optimization. The actual working concentration varies and should be decided by the user.

ELISA (For coating, order antibody without BSA)
Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at RT for 20 minutes)
Optimal dilution for a specific application should be determined.

Question

Does anti-Adipophilin / Perilipin-2 (Marker of Obesity) Monoclonal antibody M04328 work on canine IHC with erythroleukemia?

Verified Customer

Verified customer

Asked: 2019-06-10

Answer

Our lab technicians have not validated anti-Adipophilin / Perilipin-2 (Marker of Obesity) Monoclonal antibody M04328 on canine. You can run a BLAST between canine and the immunogen sequence of anti-Adipophilin / Perilipin-2 (Marker of Obesity) Monoclonal antibody M04328 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated canine samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in canine erythroleukemia in IHC, you can get your next antibody for free.
