Background of APOL1
Apolipoprotein L1 is a protein that in humans is encoded by the APOL1 gene. It is mapped to 22q12.3. This gene encodes a secreted high density lipoprotein which binds to apolipoprotein A-I. Apolipoprotein A-I is a relatively abundant plasma protein and is the major apoprotein of HDL. It is involved in the formation of most cholesteryl esters in plasma and also promotes efflux of cholesterol from cells. This apolipoprotein L family member may play a role in lipid exchange and transport throughout the body, as well as in reverse cholesterol transport from peripheral cells to the liver. Several different transcript variants encoding different isoforms have been found for this gene.

Western blot analysis of APOL1 using anti-APOL1 antibody (A01841-1).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human plasma tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-APOL1 antigen affinity purified polyclonal antibody (Catalog # A01841-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for APOL1 at approximately 37KD. The expected band size for APOL1 is at 44KD.

IHC analysis of APOL1 using anti-APOL1 antibody (A01841-1).
APOL1 was detected in paraffin-embedded section of human liver cancer tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml rabbit anti-APOL1 Antibody (A01841-1) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-Apolipoprotein L1/APOL1 antibody A01841-1 for human tissue, and we are content with the ELISA results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on goat tissues as well?

The anti-Apolipoprotein L1/APOL1 antibody (A01841-1) has not been tested for cross reactivity specifically with goat tissues, but there is a good chance of cross reactivity.
