Background of Carbonic Anhydrase XII/CA12
Carbonic anhydrase 12 is an enzyme that in humans is encoded by the CA12 gene. It is mapped to 15q22.2. Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. This gene product is a type I membrane protein that is highly expressed in normal tissues, such as kidney, colon and pancreas, and has been found to be overexpressed in 10% of clear cell renal carcinomas. Three transcript variants encoding different isoforms have been identified for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human PC-3 whole cell lysates,
Lane 3: human MCF-7 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CA12 antigen affinity purified polyclonal antibody (Catalog # A04063-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CA12 at approximately 45 kDa. The expected band size for CA12 is at 39 kDa.

We are currently using anti-CA12 antibody A04063-1 for human tissue, and we are well pleased with the ELISA results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on horse tissues as well?

The anti-CA12 antibody (A04063-1) has not been tested for cross reactivity specifically with horse tissues, though there is a good chance of cross reactivity.
