20 Customer Q&As for Anti-Collagen I/COL1A1 Antibody

Question

Please see the customer's message. Which antibody do you recommend for their application? "I am conflicted about which primary antibody to order. We are planning to do a Western Blot to measure Collagen Type I in our cell lines (keratinocytes, human mesenchymal stem cells, human dermal fibroblasts cells, and dermal microvascular endothelial cells). I see that the "Anti-Collagen I Rabbit Monoclonal Antibody" detects the Collagen alpha-2 (COL1A2) chain while the other Collagen Type I antibodies you offer detect the Collagen alpha-1 (COL1A1) chain. I am conflicted about which of these two would be best for my studies. I would like advice and more background on the difference between these two and what would be most appropriate for me."

Asked: 2020-04-29

Answer

There was no obvious difference between COL1A1 and COL1A2 in terms of expression level and location in the samples. Please recommend the customer to use PA2140-2 on the basis of our test result. It is more suitable for working on the inquired samples.

Question

Will PA2140-2 anti-Collagen I/COL1A1 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Verified Customer

Verified customer

Asked: 2020-02-25

Answer

It shows on the product datasheet, PA2140-2 anti-Collagen I/COL1A1 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Question

Our team were well pleased with the WB result of your anti-Collagen I/COL1A1 antibody. However we have observed positive staining in brain secreted using this antibody. Is that expected? Could you tell me where is COL1A1 supposed to be expressed?

G. Krishna

Verified customer

Asked: 2016-01-29

Answer

From literature, brain does express COL1A1. Generally COL1A1 expresses in secreted, extracellular space, extracellular. Regarding which tissues have COL1A1 expression, here are a few articles citing expression in various tissues:
Bone, Pubmed ID: 3340531
Brain, Pubmed ID: 15489334

Verified Customer

Verified customer

Asked: 2016-01-19

Answer

Our lab technicains tested tissue samples which were treated with lysis buffer. C-propeptide in the protein present in the samples was cleaved. We haven't worked on samples containing protein with an intact C-propeptide. We are uncertain if PA2140-2 would recognize the epitope in the presence of an intact C-propeptide.

Question

Does anti-Collagen I/COL1A1 antibody PA2140-2 work on monkey WB with spinal cord?

R. Johnson

Verified customer

Asked: 2015-11-09

Answer

Our lab technicians have not validated anti-Collagen I/COL1A1 antibody PA2140-2 on monkey. You can run a BLAST between monkey and the immunogen sequence of anti-Collagen I/COL1A1 antibody PA2140-2 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated monkey samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in monkey spinal cord in WB, you can get your next antibody for free.

Question

My question regarding product PA2140-2, anti-Collagen I/COL1A1 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

N. Krishna

Verified customer

Asked: 2015-11-09

Answer

We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PA2140-2 anti-Collagen I/COL1A1 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
