Background of DC-LAMP
APC11 is also known as Anaphase promoting complex subunit 11, APC11, Cyclosome subunit 11, Hepatocellular carcinoma associated RING finger protein, and HSPC214. APC11 is a component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC11 may function to recruit the E2 ubiquitin-conjugating enzymes to the complex. APC11 interacts with the cullin domain of ANAPC2 and also interacts with UBE2D2. APC11 shows both a cytoplasmic and nuclear localization. APC11 is expressed at high levels in skeletal muscle and heart; in moderate levels in brain, kidney, and liver; and at low levels in colon, thymus, spleen, small intestine, placenta, lung and peripheral blood leukocyte. APC11 is a member of the RING-type zinc finger family and is auto-ubiquitinylated.

Would A09406 anti-DC-LAMP antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
As indicated on the product datasheet, A09406 anti-DC-LAMP antibody as been validated on IHC. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

Our lab technicians have not validated anti-DC-LAMP antibody A09406 on goat. You can run a BLAST between goat and the immunogen sequence of anti-DC-LAMP antibody A09406 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated goat samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in goat visceral pleura in IHC, you can get your next antibody for free.
