IHC analysis of IL18 using anti-IL18 antibody (PB9014).
IL18 was detected in a paraffin-embedded section of mouse lung tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-IL18 Antibody (PB9014) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

Background of Il18
Interleukin (IL)-18, also called Interferon-gamma-inducing factor (IGIF), augments natural killer (NK) activity in spleen cells. The gene encodes a precursor protein of 192 amino acids and a mature protein of 157 amino acids. IL18 is a recently discovered cytokine that modulates both T helper type 1 (Th1) and Th2 responses. IL18 is a potent proinflammatory cytokine with potential atherogenic properties. It is highly expressed in the atherosclerotic plaques compared with control normal arteries and is localized mainly in plaque macrophages.
