Background of MPP2
MAGUK p55 subfamily member 2 is a protein that in humans is encoded by the MPP2 gene. Palmitoylated membrane protein 2 is a member of a family of membrane-associated proteins termed MAGUKs (membrane-associated guanylate kinase homologs). MAGUKs interact with the cytoskeleton and regulate cell proliferation, signaling pathways, and intracellular junctions. Palmitoylated membrane protein 2 contains a conserved sequence, called the SH3 (src homology 3) motif, found in several other proteins that associate with the cytoskeleton and are suspected to play important roles in signal transduction.

Western blot analysis of MPP2 expression in rat brain extract (lane 1) and mouse brain extract (lane 2). MPP2 at 65KD was detected using rabbit anti-MPP2 Antigen Affinity purified polyclonal antibody (Catalog # PB10079) at 0.5 μg/mL. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

Overlay histogram showing A431 cells stained with PB10079 (Blue line).The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-MPP2 Antibody (PB10079,1μg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (BA1127, 5-10μg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1μg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

We are currently using anti-MPP2 antibody PB10079 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on bovine tissues as well?

The anti-MPP2 antibody (PB10079) has not been validated for cross reactivity specifically with bovine tissues, but there is a good chance of cross reactivity. It shows on the product datasheet, PB10079 anti-MPP2 antibody as been tested on Flow Cytometry. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
