Background of PRDX6
PRDX6 is also known as PRX, p29 or AOP2. The protein encoded by this gene is a member of the thiol-specific antioxidant protein family. This protein is a bifunctional enzyme with two distinct active sites. It is involved in redox regulation of the cell; it can reduce H (2)O (2) and short chain organic, fatty acid, and phospholipid hydroperoxides. It may play a role in the regulation of phospholipid turnover as well as in protection against oxidative injury.

Western blot analysis of PRDX6 using anti-PRDX6 antibody (PB9350).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human Jurkat whole cell lysates,
Lane 3: human MCF-7 whole cell lysates,
Lane 4: human 293T whole cell lysates,
Lane 5: rat lung tissue lysates,
Lane 6: rat liver tissue lysates,
Lane 7: mouse lung tissue lysates,
Lane 8: mouse liver tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PRDX6 antigen affinity purified polyclonal antibody (Catalog # PB9350) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PRDX6 at approximately 25 kDa. The expected band size for PRDX6 is at 25 kDa.

ICC analysis of PRDX6 using anti-PRDX6 antibody (PB9350).
PRDX6 was detected in immunocytochemical section of PC-3 cell. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (AR0022) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 1μg/ml rabbit anti-PRDX6 Antibody (PB9350) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The section was developed using Strepavidin-Biotin-Complex (SABC)(Catalog # SA1022) with DAB as the chromogen.

We are currently using anti-Peroxiredoxin 6/PRDX6 antibody PB9350 for human tissue, and we are content with the ICC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it true that the antibody can work on pig tissues as well?

The anti-Peroxiredoxin 6/PRDX6 antibody (PB9350) has not been validated for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity.
