Western blot analysis of PRKCB using anti-PRKCB antibody (M01940).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human K562 whole cell lysates,
Lane 2: rat brain tissue lysates,
Lane 3: mouse brain tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PRKCB antigen affinity purified monoclonal antibody (Catalog # M01940) at 1:500 overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:500 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PRKCB at approximately 77 kDa. The expected band size for PRKCB is at 77 kDa.

Iodixanol triggered PKC phosphorylation, NOX activation, and eNOS dysregulation in HUVECs. HUVECs were incubated with iodixanol (30 mg I/ml) for 0, 5, 15, 30, 60, or 120 min. The phosphorylation of PKC beta II, p47, Rac1, eNOS (Thr495), and eNOS (Ser1177) was assayed by Western blot analysis (A-E) .

D-4F inhibited iodixanol-induced NOX activation and eNOS dysregulation through the AMPK/PKC pathway in HUVECs. HUVECs were preincubated in the absence or presence of Compound C (2 μmol/L) for 1 h. Then, cells were incubated with or without D-4F (20 μg/ml) for 8 h and treated with iodixanol (30 mg I/ml) for 30 min. The phosphorylation of PKC beta II, p47, Rac1, and eNOS (Thr495) was assayed by Western blot analysis (A-D) .

You can run a BLAST between zebrafish and the immunogen sequence of anti-PKC beta 2 Rabbit Monoclonal antibody M01940 to see if they may cross-react. As indicated on the product datasheet, M01940 anti-PKC beta 2 Rabbit Monoclonal antibody as been validated on IP. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
