Background of TRPV4
TRPV4 (Transient Receptor Potential Cation Channel Subfamily V Member 4), also known as TRP12, OTRPC4 or VROAC, is a protein that in humans is encoded by the TRPV4 gene. This gene encodes TRPV4, a member of the OSM9-like transient receptor potential channel (OTRPC) subfamily in the transient receptor potential (TRP) superfamily of ion channels. By genomic sequence analysis, Liedtke et al. (2000) mapped the TRPV4 gene to chromosome 12q24.1. They mapped the mouse Trpv4 gene to distal chromosome 5 by radiation hybrid analysis. Wissenbach et al. (2000) found that hypoosmotic conditions rapidly activated TRP12, while hyperosmotic conditions inhibited the activity.Strotmann et al. (2000) showed that OTRPC4 was responsive to changes in extracellular osmolarity in the physiologically relevant range and was expressed in tissues exposed to changing osmotic environments.

Western blot analysis of TRPV4 using anti-TRPV4 antibody (A00565-2).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HEK293 whole cell lysates,
Lane 2: rat PC-12 whole cell lysates,
Lane 3: rat C6 whole cell lysates,
Lane 4: mouse brain tissue lysates,
Lane 5: mouse testis tissue lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TRPV4 antigen affinity purified polyclonal antibody (Catalog # A00565-2) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TRPV4 at approximately 98KD. The expected band size for TRPV4 is at 98KD.
