Background of Versican

Versican (VCAN), also known as CSPG2, is a large extracellular matrix proteoglycan that is present in a variety of human tissues. Versican is a large chondroitin sulfate proteoglycan with an apparent molecular mass of more than 1000kDa. The Versican gene is mapped on 5q14.2-q14.3. The human versican gene contains 15 exons spanning more than 90 kb. The distribution of versican by using affinity-purified polyclonal antibodies that recognize the core protein of the prominent versican splice variants V0 and V1. Versican staining was noted in the central and peripheral nervous system, in the basal layer of the epidermis, and on the luminal surface of some glandular epithelia. Biochemical purification of LLC-conditioned medium led to identification of the extracellular matrix proteoglycan versican, which is upregulated in many human tumors including lung cancer, as a macrophage activator that acts through TLR2 and its coreceptors TLR6 and CD14. By activating TLR2:TLR6 complexes and inducing TNF-alpha secretion by myeloid cells, versican strongly enhances LLC metastatic growth.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Working Antibody for Versican--Gang Ye, BIOLOGY, YORK UNIVERSITY, RESEARCH ASSOCIATE/LAB MANAGER

Detection ECL

"Since versican has several isoforms, the current antibodies targeting versican on the market is very tricky. Most of them didn't give us a reasonable result. Some of them only detect a band at 70 KD, which is unlikely a potent component of versican protein. Bosterbio claims this antibody dectects a band over 250 KD. Since versican is a very big protein. It sounds this is a real antibody. After testing, we found we can see a clear detected band around 245 KD. We realized that this might be the true result...Although 245 KD is still smaller than the versican core protein, it is likely the true result of versican dectection."

Question

Can you help my question with product PB9453, anti-Versican/VCAN antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

R. Yang

Verified customer

Asked: 2019-06-04

Answer

We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free PB9453 anti-Versican/VCAN antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.

Question

Will anti-Versican/VCAN antibody PB9453 work on zebrafish IHC with glial tumor?

P. Li

Verified customer

Asked: 2015-11-27

Answer

Our lab technicians have not tested anti-Versican/VCAN antibody PB9453 on zebrafish. You can run a BLAST between zebrafish and the immunogen sequence of anti-Versican/VCAN antibody PB9453 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated zebrafish samples, this use of the antibody is not covered by our guarantee. Does PB9453 anti-Versican/VCAN antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

A. Jackson

Verified customer

Asked: 2013-05-30

Answer

It shows on the product datasheet, PB9453 anti-Versican/VCAN antibody as been tested on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
