The RAB9 gene encodes the Ras-related protein Rab-9A. The protein maps to Xp22.2 and has been implicated in the recycling of membrane receptors. In HeLa cells, downregulation of RAB9A gene expression results in severe vacuolation. In addition, the RAB9A GTPase directly binds TIP47 in the active GTP-bound conformation, enhancing the affinity of the protein cargo. Furthermore, RAB9A interacts with TIP47 and RABEPK.
Rab proteins are master regulators of intracellular vesicle transport and regulate the recruitment of specific regulators and effectors. Rabs are involved in vesicle budding, tethering to the membrane, and mobility through interactions with the cytoskeleton. Rabs are known to be targets for multiple diseases, but their function in the kidney is not well understood. Rab3A-Rph3A complex are relevant to cell morphology, cell loss, and fifiltration function in the kidney.
In many cases, the expression of the RAB9A marker is used to identify late endosomes. Rab9A is located in late endosomes and is involved in retrograde transport of cargo from late endosomes to the TGN. Rab11a localizes to recycling endosomes, which recycle cargo from early endosomes back to the plasma membrane. These are two important functions of the RAB9A protein.
The RAB9A gene encodes the endocytic protein GOLM1. The Y22A mutation affects a critical endocytic signal, resulting in a significant deficiency in removal from the plasma membrane. Y22A colocalizes with Rab5a and is present in ten percent of Rab5-positive vesicles. Moreover, Rab9A is essential for the recycling of the env protein.
The RAB9A marker is highly specific for RAB9A, a member of the Ras superfamily of small GTPases. Rabs help in intracellular protein transport, and different members of the family dock and fusion vesicles. Rab 9 facilitates the transport of the mannose 6-phosphate receptor. Rab 9 also promotes recycling of vesicles. Rab9A is prenylated by two geranylgeranylgeranyl groups at its C-terminus.
Another advantage of using the RAB9A marker is that it can be used to detect mCherryTM-tagged endosomes. Rab9a colocalizes with mCherry tagged recycling endosomes. mCherryTM variants of the RAB9A marker were found to colocalize with the RAB9a signal in about 2% of the Rab11a-positive endosomes.
The validity of the RAB9A marker in BoSTER Bio assays has been established through a series of studies. The antibody is a monoclonal antibody to the protein TYRP1, which reacts with TYRP1 in a variety of animal samples. Boster Bio developed its antibody to TYRP1 using rabbit and mouse. The RAB9A protein catalyzes the oxidation of DHICA to indole-5,6-quinone-2-carboxylic acid (INQ2C), a signal that indicates the presence of RAB9A protein.
Available in several catalogs, the anti-RAB9A antibody by Boster Bio reacts with Human, Mouse and Rat. It has been validated on Western Blotting and Immunohistochemistry, so you can trust its validity. Boster Bio is also a member of the prestigious Molecular Probes network, which provides the most comprehensive array of primary antibodies available in the market.
