Background of NR3C1

The glucocorticoid receptor (GR, or GCR), also known as NR3C1, is the receptor to which cortisol and other glucocorticoids bind. In humans, the GR protein is encoded by NR3C1 gene which is located on chromosome 5 (5q31). GR is expressed in almost every cell in the body and regulates genes controlling the development, metabolism, and immune response. Because the receptor gene is expressed in several forms, it has many different (pleiotropic) effects in different parts of the body. The activated GR complex up-regulates the expression of anti-inflammatory proteins in the nucleus or represses the expression of pro-inflammatory proteins in the cytosol (by preventing the translocation of other transcription factors from the cytosol into the nucleus).

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

2 Customer Q&As for Anti-Glucocorticoid Receptor/NR3C1 Antibody Picoband™

Question

What cell lysis protocol was used for the cell samples of PB9342?

Verified customer

Asked: 2019-12-17

Answer

For the Anti-Glucocorticoid Receptor/NR3C1 Antibody Picoband™ (PB9342), please see the following cell lysis protocol. 1. Rinse cells in ice-cold PBS. 2. Scrape cells or trypsinize cells and wash with PBS. 3. Centrifuge at ~3000 x g for 2-3 minutes, remove PBS. The centrifugation force and time can be varied depending on the cell type. 4. Add lysis buffer. 5. Sonicate for 10–15 sec to complete cell lysis. 6. Centrifuge at ~10000 x g for 10 minutes. The centrifugation force and time can be varied depending on the cell type. 7. Aspirate supernatant avoiding pellet and upper, lipid-containing phase. 8. Aliquot supernatant and store at -20°C.

Asked: 2019-10-16

Answer

The anti-Glucocorticoid Receptor/NR3C1 antibody (PB9342) has not been validated for cross reactivity specifically with canine tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in canine you can get your next antibody for free. Please contact me if I can help you with anything.
