Background of Angiopoietin-1

Angiopoietin 1, also called ANG1 is a type of angiopoietin and is encoded by the gene ANGPT1. Angiopoietins are proteins with important roles in vascular development and angiogenesis. All angiopoietins bind with similar affinity to an endothelial cell-specific tyrosine-protein kinase receptor. This gene was mapped to 8q23.1. The protein encoded by this gene is a secreted glycoprotein that activates the receptor by inducing its tyrosine phosphorylation. It plays a critical role in mediating reciprocal interactions between the endothelium and surrounding matrix and mesenchyme and inhibits endothelial permeability. The protein also contributes to blood vessel maturation and stability, and may be involved in early development of the heart.

A: do not freeze and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test Angiopoietin-1 for a later time, you should let the blood clot in glass tubes and separate the serum to freeze for later analysis.

Below is our sample preparation protocol for cell lysate for the Mouse Angiopoietin-1 ELISA Kit PicoKine™ (EK1296). Cell Lysate Sample Preparation: Collect and rinse cells in PBS. Homogenize and lyse cells thoroughly in lysis buffer (e.g. Mammal Cell Protein Extraction Reagent, Boster Catalog Number: AR0103). Centrifuge cell lysate at approximately 10,000 x g and 4°C for 5 min. Assay immediately or aliquot supernatant and hold at -80°C (Avoid freeze/thaw cycles). Guidelines on lysis buffer: Avoid more than 0.1% SDS or other strongly denaturing detergents (In general, non-ionic detergents such as Triton X-100 or NP-40 are best. Zwitterionic detergents such as CHAPS or mild ionic detergents such as sodium deoxycholate will work). Do not use more than 2% v/v total detergent. Avoid the use of sodium azide. Avoid more than 10 mM reducing agents (e.g. dithiothreitol, mercaptoethanols).

A: unable to know the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the Angiopoietin-1 ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.

A: Chelating agents such as EDTA, Heparin and Citrate can sequester metal ions from the functional domain of Angiopoietin-1 causing degradation of its protein structure. Angiopoietin-1 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes needed for detection and block the antigen antibody reaction. We have tested the Angiopoietin-1 ELISA, treating samples with various anticoagulants and decided that heparin or EDTA can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

We have not tested the Mouse Angiopoietin-1 ELISA Kit PicoKine™ (EK1296) with tissue lysates/homogenates before. For tissue homogenates, endogenous biotin should be considered. Endogenous biotin in tissue homogenates might introduce false positive signal. Please run the test as described below to confirm if there is Endogenous biotin in the tissue lysate samples using the ELISA kit. • Add tissue homogenates into the wells and then add ABC and TMB without adding any biotinylated detection antibody to see if any signal will be observed. • If no signal is produced, then you can work on the tissue sample by using the kit.
