Background of HSPA6
Frequenin, or Neuronal calcium sensor, is a regulator of G protein-coupled receptor phosphorylation in a calcium dependent manner. It directly regulates GRK1 (RHOK), but not GRK2 to GRK5. It can substitute for calmodulin and it stimulates PI4KB kinase activity. It is involved in long-term synaptic plasticity through its interaction with PICK1. Ncs1 may also play a role in neuron differentiation through inhibition of the activity of N-type voltage-gated calcium channel.

Western blot analysis of HSP70B' expression in untreated HELA whole cell lysates (lane 1), HELA (heat shocked 2 hours) cell lysates (lane 2) and HSP70B' protein (lane 3). HSP70B' at 70KD was detected using rabbit anti-HSP70B' Antigen Affinity purified polyclonal antibody (Catalog # A05402) at 1 μg/mL. The blot was developed using chemiluminescence (ECL) method (Catalog # EK1002).

We are currently using anti-HSP70B' antibody A05402 for human tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on pig tissues as well?

The anti-HSP70B' antibody (A05402) has not been tested for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity. Will A05402 anti-HSP70B' antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
It shows on the product datasheet, A05402 anti-HSP70B' antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
