Storage & Handling

Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use. Expiration date is one (1) year from date of opening.

Background of NEK2
NEK2, also called serine/threonine-protein kinase Nek2NimA-related protein kinase 2, NimA-like protein kinase 1 and HSPK 21, is a protein kinase involved in mitotic regulation that may have a role at the G2-M transition and may also play a role in meiosis. NEK2 has a nuclear localization and accumulates throughout S phase and shows maximal levels in late G2. This expression pattern is highly reminiscent of that of A and B cyclins.

Western blot analysis of NEK2 expression in A-20 whole cell lysates (lane 3). NEK2 at 52KD was detected using rabbit anti-NEK2 Antigen Affinity purified polyclonal antibody (Catalog # A01606) at 1:500. The blot was incubated with a 1:500 dilution of the antibody at room temperature followed by detection using standard techniques. Personal communication Steven Pelech, Kinexus Inc.

We are currently using anti-NEK2 antibody A01606 for human tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on bovine tissues as well?

The anti-NEK2 antibody (A01606) has not been validated for cross reactivity specifically with bovine tissues, though there is a good chance of cross reactivity. Our lab technicians have not tested anti-NEK2 antibody A01606 on feline. You can run a BLAST between feline and the immunogen sequence of anti-NEK2 antibody A01606 to see if they may cross-react. If the sequence homology is close, then you can perform a pilot test. Keep in mind that since we have not validated feline samples, this use of the antibody is not covered by our guarantee. However we have an innovator award program that if you test this antibody and show it works in feline lymphoblast in WB, you can get your next antibody for free.

Question
My question regarding product A01606, anti-NEK2 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

Answer
We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free A01606 anti-NEK2 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
