Background of IL-6

Interleukin-6 (IL-6) is a protein that in humans is encoded by the IL6 gene. Interleukin-6 (IL-6) is involved not only in the hepatic acute phase response but also in adipose tissue metabolism, lipoprotein lipase activity, and hepatic triglyceride secretion. IL-6 is a pleiotropic cytokine that plays a critical role in bone resorption. IL-6 has been suggested to stimulate the HPA axis during immune activation independent of the input of hypothalamic corticotropin-releasing hormone (CRH). Interleukin-6 (IL-6) is an immunoregulatory cytokine that activates a cell-surface signaling assembly composed of IL-6, the IL-6 alpha-receptor (IL-6Ralpha), and the shared signaling receptor gp130. The standard product used in this kit is recombinant rat IL-6, consisting of 188 amino acids with the molecular mass of 21.8kDa.

10 Customer Q&As for Rat IL-6/Interleukin-6 ELISA Kit PicoKine®

Question

Q: which procedure should I follow in order to thaw whole blood sample for IL-6 ELISA after freezing?

Verified Customer

Verified customer

Asked: 2020-05-10

Answer

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-6 for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.

Question

I would like to know more about Rat IL-6 ELISA Kit antibody. Could this be used also as neutralizing antibody? As it stated in the product notes that this antibody can be used to block IL-6/IL-6R interactions. You think it is just the same as neutralizing antibody?

Verified Customer

Verified customer

Asked: 2020-03-19

Answer

The Rat IL-6 ELISA Kit antibody can be used to prevent IL-6 binding to its receptor, therefore it could be used in a neutralizing type of assay.

Question

To Technical Support: I would like to inquire if this Rat IL-6 ELISA Kit can be used for urine samples?

Verified Customer

Verified customer

Asked: 2020-01-08

Answer

The Rat IL-6 ELISA Kit has been successfully tested with urine samples, we can guarantee that it will worked well. However, we would recommend to start a minimal dilution (such as 2-fold) since based on our experience cytokine levels are often very low in urine. Here

Question

Q: can I use citrate plasma as samples in Rat IL-6 Picokine® ELISA Kit (Catalog # EK0412)?

Verified Customer

Verified customer

Asked: 2019-03-18

Answer

A: Chelating agents such as EDTA, Heparin and Citrate can irreversibly bind metal ions from the functional domain of IL-6 causing degradation of its protein structure. IL-6 may be denatured as a result and may compromise the assay's measurements. The chilating sites could also be too close to the epitopes required for detection and block the antigen antibody reaction. We have tested the IL-6 ELISA, treating samples with various anticoagulants and decided that heparin, EDTA or citrate can be used for treatment of blood/plasma samples. Do not use other anticoagulents when collecting samples.

Question

Q: how many samples can be assayed in a Picokine® ELISA Kit?

Verified Customer

Verified customer

Asked: 2019-02-25

Answer

A: The Picokine® ELISA Kits will generally run a 7-point standard curve, non-specific binding wells, and 39 samples in duplicate. this may might differ by kit so please refer to each datasheet for details.

Question

Can EK0412 be used with cytosolic fractions?

Verified customer

Asked: 2019-02-08

Answer

We haven't tested tissue lysates/cytosolic fractions with Rat IL-6 ELISA Kit PicoKine EK0412. Endogenous biotin in tissue homogenates might introduce false positive signal. We recommend to run a pilot test to confirm the effect of endogeous biotin first. Add tissue homogenates into the wells then add ABC and TMB without adding any biotinylated detection antibody to see if any signal will be observed. If no signal is produced, proceed and work on the tissue sample by using the kit.

Question

Q: how are cell lysates prepared for use in Picokine® ELISA kits?

Verified Customer

Verified customer

Asked: 2018-11-16

Answer

A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are included in the product insert. Components in lysate and lysis buffer can have an affect on immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question

Q: What is the optimal O.D. value for IL-6 ELISA kit? I performed your IL-6 ELISA on serum samples. For my positive control, I received an O.D. value of 0.826, while my negative control received a value of 0.136. I obtained both of these controls from the ELISA kit, where your kit's typical data shows O.D. values much higher than my positive control and your background is lower. My samples O.D. values are around 0.225 and the highest is only 0.357. can I consider these samples contain IL-6 even though the O.D. values are not very high?

Verified Customer

Verified customer

Asked: 2015-04-07

Answer

A: The absolute O.D. values may change according to incubation time. The more you incubate the higher the O.D. values are going to be. a point of focus should be is whether your sample O.D. values are statistically significantly higher than your blank values. regarding your protocol, you could extend your development time in the substrate incubation step to obtain higher O.D. values, as long as your negative controls' O.D. values are not increasing faster in relation to your positive controls. normally, a sample with O.D. value 2 standard deviations higher than your negative controls can be considered positive. We calculate the sensitivity of this ELISA kit by converting cutoff O.D. value, calculated as the average of 20 negative controls plus 2 standard deviations of the 20 negative controls, into a concentration. in other words, when we claim this IL-6 ELISA kit to have sensitivity of 5pg/ml, that means the minimum amount of IL-6 that can be declared/interpreted as positive by the above standard is 5pg/ml.
