Background of Ccl2

Monocyte chemotactic protein-1 (MCP-1) is a member of the small inducible gene (SIG) family. It has been shown to play a role in the recruitment of monocytes to sites of injury and infection. By analysis of a panel of somatic cell hybrids, we have localized the MCP-1 gene, designated SCYA2, to human chromosome 17. In situ hybridization confirmed this assignment and further localized the gene to 17q11.2-q21.1. MCP-1 plays a unique and crucial role in the initiation of atherosclerosis and may provide a new therapeutic target in this disorder. Monocyte chemoattractant protein-1 (MCP-1), regulated on activation, normal T cell expressed and secreted, and stimulation of monocytes from healthy carriers of the genotype GG with Mycobacterium tuberculosis antigens yielded higher MCP-1. The standard used in this kit is recombinant mouse MCP-1 (Q24-R96) with the molecular mass of 8.5Kda.

"Had to perform the analysis twice initially as the background wasn't clear. Overall, good reproducible estimation. Reliable, reproducible. Good product but would appreciate a faster assay kit with less chances of manual error." (Customer’s Tip: "Perform washing steps at least 6-7 times to get a clearer background with 60 seconds wait period in between each washing.")

Question

Q: what is the protocol regarding preparation of cell lysates prepared for use in Picokine® ELISA kits?

Verified Customer

Verified customer

Asked: 2019-06-01

Answer

A: in those Picokine® ELISAs where cell or tissue lysate is a validated sample type, sample preparation instructions for lysate are present in the product insert. Components in lysate and lysis buffer can change immunoreactivity, so if lysate is not a validated sample type, care must be taken in sample preparation and validation.

Question

Can the sample diluent buffer, ABC diluent buffer and antibody diluent buffer be used interchangeably between the kit?

Verified customer

Asked: 2019-05-28

Answer

For the Mouse IL-1 Beta ELISA Kit PicoKine™ (EK0394), Mouse MCP-1 ELISA Kit PicoKine™ (EK0568), Mouse TNF Alpha ELISA Kit PicoKine™ (EK0527), Mouse IL-6 ELISA Kit PicoKine™ (EK0411) and Mouse Adiponectin ELISA Kit PicoKine™ (EK0596), the sample diluent buffer, ABC diluent buffer and antibody diluent buffer can be used interchangeably between the kit. However, the sample diluent buffer can only be used to dilute standards and samples, ABC diluent buffer is used to dilute ABC only and antibody diluent buffer is used to dilute antibody only.

Question

Q: Are Boster Bio recombinant proteins and antibodies sterile?

W. Wright

Verified customer

Asked: 2017-08-30

Answer

A: although the vials are bottled using aseptic techniques, heat-treated vials, and sterile stock solutions, they are not considered or guaranteed to be sterile. If sterile material is required for an experiment, the material can be filtered through a 0.2 micron filter designed for use with biological fluids.

Question

Q: if the enzyme conjugated CCL2 antibodies are mixed with the substrate, will that convert the substrate into the enzymatic reaction product? Or the enzyme function is only activated when the antibody is attached with the CCL2 antigen?

Verified Customer

Verified customer

Asked: 2014-05-22

Answer

A: The enzyme is always active. Avoid contaminating the substrate with enzyme prior to the incubation otherwise it compromises the assay with false positive signal.
