Background of MECP2
MeCP2 is a chromosomal protein with abundant binding sites in the chromatin. It belongs to the family of methyl CpG binding proteins which also comprises MBD1, MBD2, MBD3 and MBD4. MeCP2 can bind specifically to methylated promoters, thereby repressing transcription. This transcriptional repression is mediated through interaction with histone deacetylase and the corepressor SIN3A. MeCP2 also is essential for development. Mutations in MeCP2 are the cause of several types of mental retardation including Rett syndrome, a progressive neurological disorder that causes mental retardation in females and mental retardation syndromic X-linked type 13, and may also be involved in Angelman syndrome and susceptibility to some types of autism.

ChIP assays were performed using human osteosarcoma (U2OS) cells, Anti-Suz12 polyclonal antibody (Catalog # CI1047) and optimized PCR primer sets. Sheared chromatin from 1x10e6 cells and 5 μg of antibody were used per ChIP experiment. IgG (1 μg/IP) was used as a negative IP control. Quantitative PCR was performed with primers for the promoter of the ZMYND10 gene, used as a positive control and for the promoter of the CDC6 gene, used as a negative control.

To determine the titer of the antibody, an ELISA was performed using a serial dilution of Anti-MeCP2 polyclonal antibody (Catalog # CI1047) and the crude serum. The plates were coated with the peptide used for immunization of the rabbit. By plotting the absorbance against the antibody dilution, the titer of the purified antibody was estimated to be: 1:32,900.

Based on literature skeletal muscle does express MECP2. Based on Uniprot.org, MECP2 is expressed in cerebellar vermis, placenta, colon endothelium, skeletal muscle, cervix carcinoma, t-cell, leukemic t-cell, cervix carcinoma erythroleukemia, liver, among other tissues. As indicated on the product datasheet, CI1047 anti-MeCP2 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.

I have a question about product CI1047, anti-MeCP2 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?

We do not recommend storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free CI1047 anti-MeCP2 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
