Background of IL-1 beta/IL-1F2

Interleukin-1beta (IL-1beta) is a potent stimulator of bone resorption whose gene is mapped to 2q14, and has been implicated in the pathogenesis of high bone turnover and osteoporosis. IL-1beta, a prominent microglia-derived cytokine, caused oligodendrocyte death in coculture with astrocytes and microglia, but not in pure culture of oligodendrocytes alone. It also can cause nuclear export of a specific NCOR corepressor complex, resulting in derepression of a specific subset of nuclear factor-kappa-B (NFKB)-regulated genes. Furthermore, Microenvironmental IL-1beta and, to a lesser extent, IL-1alpha are required for in vivo angiogenesis and invasiveness of different tumor cells. Additional, the cooperation of IL-1beta and PDGFB induces contractile-to-synthetic phenotype modulation of human aortic smooth muscle cells in culture. Moreover, the association with disease may be explained by the biologic properties of IL-1beta, which is an important proinflammatory cytokine and a powerful inhibitor of gastric acid secretion.

"We are a pharmaceutical startup optimizing cell based drug assays where one of our endpoints was observing the differences in mature IL-1b production in the media. I have compared several kits from the most expensive versions of R&D, over eBioscience/Affymetrix to a more budget Boster's kit. The aim was to decide what is the best price for the value and quality we are getting for reliable and replicable ELISA analyses. This assay was used to test the effect of increasing concentrations of a drug on IL-1b levels (dose response). The kit reported similar data obtained from the other two kits (R&D and eBiosciences), with a slightly larger standard deviations. Results replicated the findings of the previous kits and the assay had good sensitivity/ instructions on how to perform assay and analyze data. I would use this assay in an academic lab for a project where no more than 2-3 96-well plates will be used. It should be done carefully by a scientist that has previously performed ELISA assays. There are many steps to it. It is a good quality budget option to the other leading kits on the market. I would not recommend it for a larger drug screen assay where more than 5 ELISAs kits ought to be used."

Our lab hasn't worked on Saliva samples using EK0392. Please, run a pilot test to see if the kits could work for the specific sample. In theory, if expression level of proteins in the samples falls in the assay range of the kit, the kits are able to detect the proteins in the samples.

Q: we need your recommendation regarding the dilution ratio of serum samples for detection of IL-1 beta in Human serum? I am trying to measure a multiple analytes and it requires 100ul of diluted samples for each well. We have low serum volumes so we like to dilute as much as possible.

A: unable to understand the physiological or pathological context of your samples we cannot recommend a dilution ratio without performing a pilot test with your samples. Here is how you can perform a pilot study on your own: perform a serial dilution of your samples on the IL-1 beta ELISA kit to make sure you have a linear ascending curve followed by a plateau, which signifies the samples saturating the detection limit of the kit. Then you can pick the dilution ratios from samples in the linear part of the curve as your experimental dilution ratio.

The Human IL-1 beta EK0392 Elisa Kit has not been tested on tissue homogenates. Endogenous biotin in tissue homogenates might introduce a false positive signal. We recommend to run a pilot test to confirm the effect of the endogenous biotin first. Add tissue homogenates into the wells and then add ABC and TMB without adding any biotinylated detection antibody to see if any signal will be observed. If no signal is produced, proceed and work on the tissue sample by using the kit.

Q: What is the optimal O.D. value for IL-1 beta ELISA kit? I used your IL-1 beta ELISA on serum samples. For my positive control, I received an O.D. value of 0.826, while my negative control received a value of 0.136. I obtained both of these controls from the ELISA kit, where your kit's typical data shows O.D. values much higher than my positive control and your background is lower. My samples O.D. values are around 0.225 and the highest is only 0.357. is it safe to say these samples contain IL-1 beta even though the O.D. values are not very high?

A: The absolute O.D. values may change according to incubation time. The more you incubate the higher the O.D. values are going to be. a point of focus should be is whether your sample O.D. values are statistically significantly higher than your blank values. considering your assay, you could extend your development time in the substrate incubation step to obtain higher O.D. values, as long as your negative controls' O.D. values are not increasing faster in proportion to your positive controls. typically, a sample with O.D. value 2 standard deviations higher than your negative controls can be considered positive. We calculate the sensitivity of this ELISA kit by converting cutoff O.D. value, calculated as the average of 20 negative controls plus 2 standard deviations of the 20 negative controls, into a concentration. in other words, when we claim this IL-1 beta ELISA kit to have sensitivity of 0.15pg/ml, that means the minimum amount of IL-1 beta that can be declared/interpreted as positive by the above standard is 0.15pg/ml.

For the product EK0392 you will need a 100ul of antibody for each 96 well plate. So if you are going to use all the 96 well plate, you need 9600ul (9.6ml) volume of antibody. Usually there is a loss of few micro liters while pipetting, so we suggest preparing more than what it is required.

A: we do not recommend freezing and thaw whole blood. erythrocytes are fragile and, if frozen and thawed, will undergo hemolysis rendering the samples useless. To keep your blood samples to test IL-1 beta for a later time, you should let the blood clot in glass tubes and collect the serum to freeze for later use.
