Thank you for asking; the immunogen of EK7001 Human Proinsulin ELISA kit is human insulin. It actually react with human, rat and mouse insulin as well as with proinsulin and intermediates with same affinity. It is logical since the variances between these 3 species are insignificant (different amino acids only in 5 positions). However, it also cross-react with bovine and porcine insulin. All these antigens have been tested. I would rather suggest to use an insulin standard that matches the species of the samples you are testing.

The antibody pair that we use in the EK7001 Human Proinsulin ELISA is capture antibody and detection antibody. We have not specifically tested human analogs with EK7001 Human Proinsulin ELISA but based on sequence similarity, the kit recognized these as well. However, we wouldn't guarantee it since it hasn't been specifically tested.

Answer
We generally do not recommend exact protocols for performing assays with our products EK7001 Human Proinsulin ELISA. We like to encourage researcher to have a look at our protocols in our website and use it only as guide for their assays. Because every sample is different and may require different conditions. However, this EK7001 Human Proinsulin ELISA antibody was successfully used in mammalian cell culture media in non-reduced conditions. The non-reduced SDS PAGE gel was transferred to nitrocellulose which was then blocked with 4% skim milk for 4 hours. The membrane was washed 3 x 5mins with PBS + 0.05%Tween and then labelled. It is then diluted 1/2000 in PBS for 1 hour with gentle shaking at room temperature. The membrane was washed 3 x 5min with PBS + 0.05% Tween and then labelled with a suitable secondary which was detected by colorimetric method.
